Auditory System

The auditory system involves a complex interplay of molecules that enable the detection, transduction, transmission, and processing of auditory signals. From the reception of sound waves to the perception of sound in the brain, various systems play crucial roles in each stage of auditory processing. The process of sound reception begins with sound waves entering the ear canal and reaching the eardrum (tympanic membrane). The eardrum is a thin, flexible membrane composed of proteins like collagen, which vibrates in response to sound waves. In the middle ear, the vibrations from the eardrum are transmitted to the cochlea, a fluid-filled structure in the inner ear, through three small bones called ossicles: the malleus, incus, and stapes. These ossicles are connected by joints and are primarily composed of proteins such as collagen and elastin, allowing them to efficiently transmit vibrations. The cochlea is the site of auditory transduction, where sound vibrations are converted into electrical signals via hair cells, which are the sensory receptors in the cochlea. Hair cells have specialized projections on their surface called stereocilia, composed of actin filaments and various protein complexes. When the fluid in the cochlea moves in response to sound vibrations, the stereocilia of the inner hair cells (IHCs) bend. IHCs possess mechanotransduction channels located at the tips of their hair bundles, with TRPA1 and TRP-like MET (mechanosensitive transient receptor potential-like mechanotransduction channel) channels considered potential candidates for mediating mechanotransduction. Upon hair bundle bending, these channels open, allowing the influx of calcium and potassium ions, generating a receptor potential, which leads to the release of neurotransmitters, such as glutamate, at the synapses between IHCs and auditory nerve fibres. This neurotransmitter release triggers action potentials in the auditory nerve fibres, which are then transmitted to the brain. The auditory (cochlear) nerve consists of the axons of spiral ganglion neurons located near the hair cells. These neurons transmit electrical signals from the hair cells to the auditory brainstem, where the initial processing of auditory information occurs. At the synapses between auditory nerve fibres and neurons in the brainstem, neurotransmitters are released to convey auditory information. Glutamate is the primary excitatory neurotransmitter involved in auditory synapses, binding to glutamate receptors on the postsynaptic membrane, initiating a cascade of events that generate postsynaptic electrical responses. Within the auditory brainstem, various nuclei are also involved in processing auditory information. As auditory information is relayed from the brainstem, it ascends to reach the primary auditory cortex, located in the temporal lobe. Here, neurons respond to specific frequencies and contribute to the tonotopic organization of the auditory cortex. Multiple processes underlie the remarkable plasticity of the auditory system. For instance, during development and learning, molecular signalling pathways facilitate the establishment and refinement of synaptic connections. Additionally, in response to changes in sensory input, such as hearing loss, molecular mechanisms enable the reorganization of auditory circuits to adapt to new conditions.
