Hematopoietic progenitors are a diverse group of cells in the bone marrow that give rise to various blood cell lineages. These progenitors express specific cell surface proteins that are critical for their identification, differentiation, purification, and interaction with the bone marrow microenvironment. A large variety of CD surface markers are used in studies of hematopoietic progenitors. These include: 1) CD34, a marker of hematopoietic stem and progenitor cells (HSPCs). It is expressed on multipotent progenitors, including common myeloid progenitors (CMPs) and common lymphoid progenitors (CLPs). CD34 is often used in the isolation and purification of HSPCs for transplantation and research purposes; 2) CD38, a marker of HSPCs, particularly on CLPs. It is found on multipotent progenitors regulating their proliferation and differentiation; 7) CD44, a cell adhesion molecule with a role in cell trafficking within the bone marrow and expressed on HSPCs; 8) CD135 (FLT3 or FMS-like tyrosine kinase 3), a receptor tyrosine kinase expressed on early hematopoietic progenitors, including common myeloid progenitors (CMPs) and common lymphoid progenitors (CLPs). Mutations in FLT3 are associated with certain forms of leukaemia; 9) CD49d (integrin alpha-4), part of the very late antigen-4 (VLA-4) complex, which mediates the adhesion of hematopoietic cells to bone marrow stromal cells. It plays a role in homing and retention of HSPCs in the bone marrow; 10) CXCR4 (C-X-C chemokine receptor type 4), a receptor for the chemokine CXCL12 (stromal cell-derived factor-1 or SDF-1). It is essential for the homing of HSPCs to the bone marrow and their retention within the hematopoietic niche; 11) E-selectin ligands (such as CD162), involved in the homing and extravasation of hematopoietic cells from the blood into the bone marrow or other tissues during inflammation or tissue repair; 12) CD150 (SLAM family member 1 or signalling lymphocyte activation molecule 1), expressed on a subset of HSPCs; 13) CD41 (integrin alpha-IIb), expressed on megakaryocyte-erythroid progenitors (MEPs) and involved in platelet production. It is also a marker of the megakaryocytic lineage; 14) CD71 (transferrin receptor), expressed on erythroid progenitors and involved in iron uptake, a crucial process for red blood cell development. They also participate in cell-cell interactions, adhesion, migration, and signalling events that regulate haematopoiesis, immune responses, and haematological disorders.
