Principle of Assay
The Amino Anchor Immunoprecipitation to Mass Spectrometry Kit uses the bifunctional chemistry of the Amino Anchor to prepare pure, high-quality antigens for unbiased antigen profiling via mass spectrometry (MS). This approach is particularly useful for the identification of autoantigens and cancer antigens in a sample-specific manner using only a small volume of plasma or serum (or other auto-antibody source). First, antigens are labeled with the Amino Anchor via its amine-reactive group. Antibodies (including auto-antibodies) from a single sample are bound to Protein A resin, then used to capture target antigens. After washing away unbound proteins, antigens are eluted and recaptured on TCO resin via the resin-reactive group of the Amino Anchor, resulting in antibody removal. Pure target antigens can then be eluted for use or digested with trypsin for MS analysis without the contaminating signal from overabundant antibodies. This simplified procedure generates MS-ready peptides in as little as 6 hours, is radiolabel-free, and is compatible with a range of antibody and target antigen sources.
