Principle of Assay
The Amino Anchor Intact Universal Protein Cleanup Kit uses the bifunctional chemistry of the Amino Anchor to prepare pure, high quality protein representing the entire proteome for mass spectrometry or other downstream proteomics analysis. First, proteins are labeled with the Amino Anchor via its amine-reactive group. Via the resin-reactive group of the Amino Anchor, the protein is then captured by the addition of the capture resin, allowing removal of contaminants by washing. Pure protein can is then eluted for downstream proteomics analysis. This streamlined workflow generates intact protein in just a few hours, yields unbiased and unmodified protein samples, and is compatible with most tissues and cell lysates.

Amino Anchor Intact Universal Protein Cleanup Kit workflow. After labeling proteins in lysate with the Amino Anchor, capture resin binds to protein, enabling removal of contaminants. Intact total protein can then be eluted for downstream proteomics analysis (note that trypsin steps 8-10 apply only to the Amino Anchor Peptide Universal Protein Cleanup Kit).
