Principle of Assay
The Amino Anchor Multiomics (DNA, RNA, Protein) Sample Preparation Kit uses the bifunctional chemistry of the Amino Anchor to prepare mass spectrometry (MS)-ready protein for proteomics analysis, and separately prepares DNA and RNA that are suitable for sequencing. First, proteins are labeled with the Amino Anchor via its amine-reactive group. DNA, RNA and protein (via the resin-reactive group of the Amino Anchor) are all captured by the addition of the capture resin, allowing removal of contaminants by washing. Sequencing-ready DNA and RNA are then eluted in two separate elutions. Protein can also be eluted at this point, or it can be digested with trypsin first and then eluted for MS analysis. This streamlined procedure generates omics-ready protein, DNA and RNA in as little as 6 hours, requires only a small amount of sample (as little as 0.2mg tissue), and is compatible with most tissues and cell lysates.
