Principle of Assay
The Amino Anchor Intact Universal Protein Cleanup Kit uses the bifunctional chemistry of the Amino Anchor to prepare pure, high quality peptides representing the entire proteome for mass spectrometry analysis. First, proteins are labeled with the Amino Anchor via its amine-reactive group. Via the resin-reactive group of the Amino Anchor, the protein is then captured by the addition of the capture resin, allowing removal of contaminants by washing. Pure protein can be eluted at this point, or it can be digested into peptides with trypsin first and then eluted for MS analysis. This streamlined workflow generates intact protein or MS-ready peptides in just a few hours, yields unbiased and unmodified protein samples, and is compatible with most tissues and cell lysates.
