Name
APC Labeling Kit
Description
APC belongs to the phycobiliprotein family of highly soluble and fluorescent proteins derived from cyanobacteria and eukaryotic algae. APC is an ultra-sensitive fluorescent tracer with high emission quantum yields, that emits in the near-infrared fluorescence range. It therefore avoids interference from the autofluorescence of cellular components and other biological materials. Home Algae's APC Labeling Kit is an SH-reactive labeling kit. Modified APC has maleimide groups that can conveniently conjugate with the SH groups of antibodies. The total conjugation time including incubation takes approximately 3 hours. The three kit sizes available can label up to 50µg, up to 100µg, and up to 1mg of antibody respectively.
Applications
Flow Cytometry, FRET, Immunoassay
Components
The APC Labeling Kit contains activated APC, an antibody reducing agent, an organic solvent and a sulfhydryl blocking reagent for quenching the reaction, and a filtration tube for desalting the protein.
Molecular Weight
105 kDa
Product Form
Liquid
Storage
Store in the dark at 2-8°C. Avoid prolonged exposure to light. Do not freeze!
General Notes

APC is significantly more sensitive than conventional organic fluorophores and has been used in applications such as Flow Cytometry, Homogeneous FRET assay, and Immunoassays. APC's absorbance maximum is at 650 nm, with a resolvable emission peak at 660 nm. It shows minimal fluoresence quenching.

APC won't penetrate into the cell and cause a false-positive reaction.

Non-toxic, with less chance of affecting characteristics of the sample, human body, and environment.

Sample of Protocol

1. Add the Modifier Solution into the antibody.
2. Incubate the reaction at room temperature for 30 min.
3. Add the Pre-activated R-PE and incubate for 1 hour at room temperature, with agitation and protected from light.
4. Add Quencher into the conjugation mixture and mix completely.
5. Incubate at room temperature and keep away from light for 30 min, until the conjugation is complete.
