Principle of Assay
Cellular dehydrogenases reduce WST-8 to a soluble orange formazan dye. The amount of formazan produced is directly proportional to the number of metabolically active cells.

Dose-dependent effects of mitomycin C on HeLa cells measured by Cell Counting Kit-8 (450 nm) and Cytotoxicity LDH Assay Kit-WST (490 nm). Increasing drug concentration decreases cell viability (CCK-8) and increases cytotoxicity (LDH release).
Combined use of Cell Counting Kit-8 and Cytotoxicity LDH Assay Kit-WST with the same samples. Supernatant is used to measure LDH release from dead cells (cytotoxicity), while remaining cells are analyzed for viability using CCK-8. Both assays are based on WST reduction to formazan via NADH-dependent reactions.
