Anti-His Tag Donor Beads 680 (A334424) in combination with Acceptor Beads 615 are used to design no-wash assays for studying protein-protein interactions. These assays are ideally suited for miniaturization and automation. They exhibit remarkable sensitivity, wide dynamic range and robust performance that compares advantageously with conventional methods. The assay measures the interaction occurring between two binding partners captured onto Donor Beads 680 and Acceptor Beads 615 nanoparticles. When located in close proximity, singlet oxygen, produced upon excitation of the tagged bead at 680nm, travels up to 200nm in aqueous solution and reacts with specific compounds embedded in the Acceptor Beads 615. The latter beads then emit a signal detectable at 615nm which is proportional to the extent of interaction prevailing between binding partners A and B (Figure 1). Reading at a wavelength below that used for excitation allows for the detection of high quality signals. Anti-His Tag Donor Beads 680 are conjugated with Anti-6xHis Antibodies allowing for efficient capture of 6xHis-tagged molecules (e.g. peptides, proteins, etc.).
