Principle of Assay
LDH released from damaged cells catalyzes a coupled enzymatic reaction leading to reduction of WST to a colored formazan. The intensity of the colorimetric signal correlates with cytotoxicity.

Dose-dependent effects of mitomycin C on cells measured by Cell Counting Kit-8 and Cytotoxicity LDH Assay Kit-WST. Increasing mitomycin C concentration decreases cell viability (CCK-8, 450 nm) and increases LDH release (LDH-WST, 490 nm), demonstrating complementary assessment of viability and cytotoxicity.
