Description
The Nuclear and Cytoplasmic Protein Extraction Kit is designed for the rapid and efficient isolation of highly enriched cytoplasmic and nuclear protein fractions from eukaryotic samples, including cultured cells and fresh tissues. The kit provides a simple and reliable workflow that enables separation of cytoplasmic and nuclear proteins in approximately 90 minutes. Using a sequential extraction strategy, cytoplasmic proteins are released under hypotonic conditions through selective cell membrane disruption, followed by centrifugation to pellet intact nuclei. Nuclear proteins are then extracted using a high-salt nuclear extraction reagent to ensure efficient solubilization. The resulting protein fractions are suitable for direct use in downstream applications such as Western blotting, EMSA, DNase footprinting, reporter gene analysis, and enzyme activity assays. The kit supports up to 100 samples and allows reagent volumes to be adjusted according to sample type and input amount.

Principle of Assay
The Nuclear and Cytoplasmic Protein Extraction Kit is based on sequential differential lysis and centrifugation to separate cytoplasmic and nuclear protein fractions from eukaryotic cells and tissues. Cells are first exposed to a hypotonic cytoplasmic extraction buffer that selectively disrupts the plasma membrane while maintaining nuclear integrity, resulting in release of cytoplasmic proteins. Following low-speed centrifugation, intact nuclei are pelleted and separated from the cytoplasmic fraction. Nuclear proteins are then extracted from the isolated nuclei using a high-salt nuclear extraction reagent, which solubilizes nuclear components through disruption of protein–DNA and protein–protein interactions. This stepwise extraction strategy yields highly enriched cytoplasmic and nuclear protein fractions suitable for downstream biochemical and molecular analyses.
