Principle of Assay
This kit provides a simple method for detecting RUBISCO activity in plant tissues. The principle is one molecule of ribulose-1,5-diphosphate combines with one molecule of carbon dioxide to produce two molecules of 3-phosphoglyceric acid (PGA). Glyceraldehyde-3-phosphate can be produced from PGA through the action of 3-phosphoglycerate kinase and glyceraldehyde-3-phosphate dehydrogenase, which is accompanied by NADH oxidation to produce NAD+. NADH has a characteristic absorption peak at 340 nm, while NAD+ does not. RUBISCO activity is calculated by measuring the rate of decrease in light absorption at 340 nm.
