Principle of Assay
SPiDER-ßGal is cleaved by ß-galactosidase to generate a reactive quinone methide intermediate that covalently binds to intracellular proteins. This self-immobilizing reaction produces a stable fluorescent signal, enabling sensitive detection at the single-cell level.
