Add the substrate after incubation with horseradish peroxidase and subsequent washing. Incubate for typically 5-15 minutes and then stop the enzymatic reaction by adding an acid, e.g. 0.18 M H2SO4. This will change the color from blue to yellow. Read the absorbance at 450 nm.
