Principle of Assay
Human Adenosine A1 Receptor ELISA Kit (A335883) is a 2 hour sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Adenosine A1 Receptor in serum, plasma, cell culture supernatant, cell or tissue lysate, and other biological fluids. This QuickRun format uses a pre-coated 96-well microtiter plate and optimized ready-to-use reagents to reduce assay time. Standards and samples are added to the wells, where Adenosine A1 Receptor binds to the immobilized capture antibody. Assay-specific biotinylated detection antibody and HRP-Streptavidin reagents are then added according to the optimized QuickRun workflow. These reagents form an antibody-antigen-antibody sandwich complex with the captured Adenosine A1 Receptor. TMB substrate solution is added to visualize the HRP enzymatic reaction. This produces a blue-coloured product that changes to yellow following addition of acidic stop solution. The intensity of the yellow colour is directly proportional to the amount of Adenosine A1 Receptor captured in each well. The concentration of Adenosine A1 Receptor is calculated by reading the absorbance at 450 nm and comparing the values with the standard curve.
