Principle of Assay
Human Ferritin Light Chain ELISA Kit (A335638) is a 2 hour sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Ferritin Light Chain in serum, plasma, cell culture supernatant, cell or tissue lysate, and other biological fluids. This QuickRun format uses a pre-coated 96-well microtiter plate and optimized ready-to-use reagents to reduce assay time. Standards and samples are added to the wells, where Ferritin Light Chain binds to the immobilized capture antibody. Assay-specific biotinylated detection antibody and HRP-Streptavidin reagents are then added according to the optimized QuickRun workflow. These reagents form an antibody-antigen-antibody sandwich complex with the captured Ferritin Light Chain. TMB substrate solution is added to visualize the HRP enzymatic reaction. This produces a blue-coloured product that changes to yellow following addition of acidic stop solution. The intensity of the yellow colour is directly proportional to the amount of Ferritin Light Chain captured in each well. The concentration of Ferritin Light Chain is calculated by reading the absorbance at 450 nm and comparing the values with the standard curve.
