Principle of Assay
Human IL-17F ELISA Kit (A336265) is a 2 hour sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human IL-17F in serum, plasma, cell culture supernatant, cell or tissue lysate, and other biological fluids. This QuickRun format uses a pre-coated 96-well microtiter plate and optimized ready-to-use reagents to reduce assay time. Standards and samples are added to the wells, where IL-17F binds to the immobilized capture antibody. Assay-specific biotinylated detection antibody and HRP-Streptavidin reagents are then added according to the optimized QuickRun workflow. These reagents form an antibody-antigen-antibody sandwich complex with the captured IL-17F. TMB substrate solution is added to visualize the HRP enzymatic reaction. This produces a blue-coloured product that changes to yellow following addition of acidic stop solution. The intensity of the yellow colour is directly proportional to the amount of IL-17F captured in each well. The concentration of IL-17F is calculated by reading the absorbance at 450 nm and comparing the values with the standard curve.
