Principle of Assay
Human Secretory IgA ELISA Kit (A77322) employs HRP antibody sandwich ELISA for the quantitative measurement of human Secretory IgA in serum, plasma, tissue homogenates, and other biological fluids. Capture antibody specific for Secretory IgA is pre-coated on a 96-well microtiter plate. Samples and standards are added, and Secretory IgA binds to the immobilized antibody. Detection is performed using HRP-conjugated detection antibody, which binds to a different epitope on the captured target. Following another wash, TMB substrate solution produces a blue-coloured product that changes to yellow after addition of acidic stop solution. The intensity of yellow is proportional to the amount of Secretory IgA captured, and the concentration is determined by reading O.D. at 450nm.
