Idirect ELISA

Lyme Disease Borrelia burgdorferi sensu lato IgG ELISA Kit (A341251) is an Indirect ELISA for the quantitative measurement of b. burgdorferi sensu stricto, b. afzelii, b. garinii, b. mayonii. b. spielmanii. b. bissetiae, b. bavariensis and b. valaisiana IgG in serum and plasma. . The 96-well microtiter plate has been pre-coated with IgG antigen. The samples and standards are added to the wells and incubated. After washing with wash buffer, Biotinylated detection antibody is added and incubated. After washing with wash buffer, HRP-Streptavidin is then added and incubated. Following another wash step, TMB substrate is added to visualize the HRP enzymatic reaction. TMB is catalysed by HRP to produce a blue-coloured product that changes to yellow after the addition of the acidic stop solution. The density of yellow is proportional to the amount of IgG sample captured on the plate. The concentration of IgG can then be calculated by reading the O.D. absorbance at 450nm in a microplate reader and referring to the standard curve.

This ELISA kit includes a 96-well microplate pre-coated with proprietary Epitogen Lyme antigens and supplied ready for use. The kit also contains a peroxidase-labelled anti-human IgG antibody, 10× wash buffer, secondary diluent, TMB substrate and a protective plate seal.

The assay demonstrates 100 percent sensitivity in late-stage Lyme disease, including Lyme neuroborreliosis and Lyme carditis, and shows greater than 87.8 percent sensitivity in acute Lyme disease within six weeks of infection.

This kit uses an immunogen composed of a mixture of 120 linear immunodominant epitopes, 4 flagellin subunits, 12 full-length VlsE proteins, and 8 full-length OspC antigens to detect IgG antibodies associated with established or past Borrelia infection.
