Lyme Disease Borrelia burgdorferi sensu lato IgG ELISA Kit (A341251) employs an indirect ELISA for the quantitative measurement of IgG antibodies to Borrelia burgdorferi sensu stricto, B. afzelii, B. garinii, B. mayonii, B. spielmanii, B. bissetiae, B. bavariensis, and B. valaisiana in serum and plasma. The 96-well microtiter plate is pre-coated with Borrelia antigens. Samples and standards are added to the wells and incubated, allowing specific IgG antibodies to bind to the immobilized antigens. After washing to remove unbound material, a biotinylated detection antibody is added, followed by HRP-streptavidin. Following a further wash step, TMB substrate is added to visualize the enzymatic reaction. HRP catalyzes the conversion of TMB to a blue product, which turns yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of Borrelia-specific IgG bound to the plate. Absorbance is measured at 450 nm using a microplate reader, and antibody concentrations are determined from a standard curve.
