Principle of Assay
Mouse Fibrinogen alpha Chain ELISA Kit (A336535) is a 2 hour sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of mouse Fibrinogen alpha Chain in serum, plasma, cell culture supernatant, cell or tissue lysate, and other biological fluids. This QuickRun format uses a pre-coated 96-well microtiter plate and optimized ready-to-use reagents to reduce assay time. Standards and samples are added to the wells, where Fibrinogen alpha Chain binds to the immobilized capture antibody. Assay-specific biotinylated detection antibody and HRP-Streptavidin reagents are then added according to the optimized QuickRun workflow. These reagents form an antibody-antigen-antibody sandwich complex with the captured Fibrinogen alpha Chain. TMB substrate solution is added to visualize the HRP enzymatic reaction. This produces a blue-coloured product that changes to yellow following addition of acidic stop solution. The intensity of the yellow colour is directly proportional to the amount of Fibrinogen alpha Chain captured in each well. The concentration of Fibrinogen alpha Chain is calculated by reading the absorbance at 450 nm and comparing the values with the standard curve.
