Principle of Assay
Rat Glucocorticoid Receptor ELISA Kit (A336948) is a 2 hour sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of rat Glucocorticoid Receptor in serum, plasma, cell culture supernatant, cell or tissue lysate, and other biological fluids. This QuickRun format uses a pre-coated 96-well microtiter plate and optimized ready-to-use reagents to reduce assay time. Standards and samples are added to the wells, where Glucocorticoid Receptor binds to the immobilized capture antibody. Assay-specific biotinylated detection antibody and HRP-Streptavidin reagents are then added according to the optimized QuickRun workflow. These reagents form an antibody-antigen-antibody sandwich complex with the captured Glucocorticoid Receptor. TMB substrate solution is added to visualize the HRP enzymatic reaction. This produces a blue-coloured product that changes to yellow following addition of acidic stop solution. The intensity of the yellow colour is directly proportional to the amount of Glucocorticoid Receptor captured in each well. The concentration of Glucocorticoid Receptor is calculated by reading the absorbance at 450 nm and comparing the values with the standard curve.
