Species Independent His tag ELISA Kit is a competitive Enzyme-Linked Immunosorbent Assay (cELISA) designed for the in vitro quantitative determination of species independent His tag in his-tagged proteins in mammalian and bacteria cell lysates or cell supernatant.

Species Independent His tag ELISA Kit (A334792) employs the sandwich enzyme immunoassay technique for the quantitative measurement of species independent His tag in his-tagged proteins in mammalian and bacteria cell lysates or cell supernatant. An antibody specific for His tag has been pre-coated onto a 96-well microtiter plate. The standards and test samples are added into the wells and the His tag present in each sample is bound to the wells by the immobilized antibody. Following incubation, the wells are washed and then incubated with Biotinylated Anti-His tag Antibody, which binds the captured His tag present in each well. Following incubation, unbound biotinylated detection antibody is removed by washing, and an HRP-Streptavidin conjugate is added to the wells and the microtiter plate is incubated. Following incubation and washing, TMB substrate solution is then used to visualize the HRP enzymatic reaction by catalysis to produce a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of His tag captured in each well. The concentration of His tag can then be calculated by reading the O.D. absorbance at 450nm in a microplate reader and referring to the standard curve.

This ELISA Kit contains 10X 600 µL Capture Antibodies, 100X 40 µL Competitor, 2 Lyophilized Protein Standard Vials, 50 mL Assay Diluent #1, 10X 20 mL Wash Buffer, 12 mL TMB Solution, 12 mL Stop Solution, Pre-Coated 96-Well A-Microplate (12 x 8 well strips), and 2 Plate Seals.
