Principle of Assay
HRP Conjugated Streptavidin ELISA Kit (A326620) employs the direct enzyme immunoassay technique for the quantitative measurement of universal HRP Conjugated Streptavidin in serum, plasma and other biological fluids. The 96-well microtiter plate is pre-coated with HRP Conjugated Streptavidin antigen. Test samples and standards are added to the wells and incubated. After washing, an HRP-conjugated detection antibody specific for HRP Conjugated Streptavidin is added and incubated. Following a final wash, TMB substrate solution is used to visualize the HRP enzymatic reaction by catalysis to produce a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of HRP Conjugated Streptavidin bound on the plate. The concentration of HRP Conjugated Streptavidin can then be calculated by reading the O.D. absorbance at 450nm in a microplate reader and referring to the standard curve.
