Immunofluorescent analysis of rat cortex section stained with Anti-Ankyrin 3 Antibody [2A8], at a dilution of 1:2,000, in green, and co-stained with Anti-NF-L Antibody (A85451), at a dilution of 1:5,000, in red. Nuclear DNA is visualised in blue using Hoechst staining. Following transcardial perfusion with 4% paraformaldehyde, the brain was post-fixed for 24 hours, cut to 45 µm, and free-floating sections were stained using the above antibodies. Anti-Ankyrin 3 Antibody [2A8] stains the axonal initial segments, while Anti-NF-L Antibody (A85451) labels dendrites and axons of neuronal cells.

Western blot analysis of different tissue and cell lysates using Anti-Ankyrin 3 Antibody [2A8], at a dilution of 1:2,000, in green. The lanes contain: [Lane 1] protein standards (red), [Lane 2] NIH-3T3 cells, [Lane 3] C6 cells, [Lane 4] HEK293 cells, [Lane 5] HeLa cells, [Lane 6] SH-SY5Y cells, [Lane 7] rat brain lysates, and [Lane 8] mouse brain lysates. Bands at ~190 kDa represent Ankyrin 3 splice variants. Higher molecular weight bands at 270 kDa and 480 kDa can be seen on longer exposure. Lower molecular weight bands are likely proteolytic fragments.
Immunofluorescent analysis of cortical neuron-glial cell culture from E20 rat embryos stained with Anti-alpha II Spectrin Antibody (A85352) at a dilution of 1:500 (green) and costained with Anti-Ankyrin 3 Antibody [2A8] (A270548) at a dilution of 1:2,000 (red). Nuclei were stained with Hoechst (blue). The spectrin antibody stains submembranous cytoskeleton of neuronal cells. The Ankyrin3 antibody labels axonal initial segments of neurons.

Counterstained with Hematoxylin (blue). Note: this antibody performs well in testing with both 4% PFA and standard NBF fixed tissues but does not stain long term NBF fixed tissue effectively.
