Freshly ejaculated human sperms were washed in PBS and extracted in 3% acetic acid, 10% glycerol, 30 mM benzaminidine. The acid extract was dialyzed against 0.2% acetic acid and subsequently used for immunization.

Western blotting analysis of human clusterin using Anti-Clusterin Antibody [Hs-3] (A86810) on lysates of MCF-7 cell line and MOLT-4 cell line (clusterin non-expressing cell line; negative control) under non-reducing and reducing conditions. Nitrocellulose membrane was probed with 2 µg/ml of mouse anti-clusterin monoclonal antibody followed by IRDye800-conjugated anti-mouse secondary antibody. Clusterin was detected around 40 kDa.
