Western Blot - Anti-CRACR2A Antibody (A89954)

Western blot analysis of extracts of various cell lines, using Anti-CRACR2A Antibody (A89954) at 1:3,000 dilution. The secondary antibody was Goat Anti-Rabbit IgG H&L Antibody (HRP) at 1:10,000 dilution. Lysates/proteins were present at 25µg per lane. The blocking buffer used was 3% non-fat dry milk in TBST. Detection was with a ECL Basic Kit. Exposure time: 90s.

Experimental Conditions: IF: 4% PFA, RT 10 min; 0.1% Triton-X, RT 10 min; primary PBS 1:300, secondary PBS 1:300.

Dilution: IF: 1:300

Results Summary: Uncharacteristic cell edge staining, intense background; green stain appeared like 'sea of green algae'; should have clustered around nuclei.

Comments: High background, poor specificity; analysis on ImageJ did not improve results.
