Western blot confirmation of Nociceptin/Orphanin FQ Receptor (OPRL) in transfected HEK293 cells. Lysates from native HEK293 cells (MOCK) or cells stably expressing OPRL were probed with phosphorylation-independent anti-Nociceptin Receptor antibody (A334517) at 1:1000.
Dose-dependent phosphorylation analysis of Nociceptin/Orphanin FQ Receptor (OPRL) in OPRL-eGFP knock-in mice. OPRL-eGFP mice were injected intraperitoneally with 0.9% NaCl or 0.3 to 30 mg/kg AT-202 for 30 min. Brain homogenates were immunoprecipitated with anti-GFP agarose beads and immunoblotted with anti-OPRL (phospho Ser351) or anti-OPRL (phospho Thr362 + Ser363) antibodies at 1:1000 (upper panels). The blot was stripped and reprobed with phosphorylation-independent anti-OPRL antibody (A334517) at 1:1000 to confirm equal loading (lower panel).
