Western Blot - Anti-SARS-CoV-2 Spike Glycoprotein RBD Antibody [2G1] (A270550)
Left Panel: HEK293 cells were transfected with DNA encoding the SARS-CoV-2 Spike Glycoprotein RBD for ACE2 which had been inserted into pCI-Neo-Mod or pCI-Neo-GFP eukaryotic expression vectors, such that the cells express either the insert only or the insert fused with GFP. The lanes contain: [Lane 1] protein standard, [Lane 2] a crude homogenate of untransfected control cells, [Lane 3] a homogenate of cells expressing SARS-CoV-2 Spike Glycoprotein RBD, and [Lane 4] a homogenate of cells expressing GFP-SARS-CoV-2 Spike Glycoprotein RBD fusion. Anti-SARS-CoV-2 Spike Glycoprotein RBD Antibody [2G1] (A270550), at a dilution of 1:3,000, in green, produces clean and specific staining of bands of the expected size. The blot was co-stained with Anti-HSP60 Antibody (A85437), at a dilution of 1:5,000, in red. Right Panel: Blot of full-length recombinant SARS-CoV-2 Spike Glycoprotein expressed in HEK293 cells; [Lanes 1] protein standard, [Lane 6] shows a loading of 1 µg, and [Lane 7] shows a loading of 100 ng. On longer exposure of the blot the antibody could readily detect 10ng of the S-protein.

The nuclei of transfected and untransfected cells are shown in blue with DAPI DNA stain.

Vero cells are a cell line derived derived from monkey kidney and express an ACE2 receptor similar to that found in humans, and are therefore susceptible to infection by patient derived SARS-CoV2 virus. The cells were grown in culture, fixed, and stained with the DNA dye DAPI, revealing cell nuclei (control, top left), The remaining three panels show staining of similar cells following infection with SARS-CoV2 virus. Cells were then reacted with the cell binding domain of the SARS-CoV2 spike protein, namely Anti-SARS-CoV-2 Spike Glycoprotein RBD Antibody [2G1] (A270550), Anti-SARS-CoV-2 Spike Glycoprotein RBD Antibody [5G8] (A270557) and Anti-SARS-CoV-2 Spike Glycoprotein RBD Antibody (A270592) as indicated. Antibody binding was revealed with appropriate ALEXA green (top right, bottom left) or red (bottom right) secondary antibodies. The virus accumulates in the cytoplasm of infected cells.
