Experimental Conditions: WB: Human cell line lysate prepared using Co-IP lysis buffer (20 mM HEPES pH 8.0, 150 mM NaCl, 0.2% NP-40, 10% glycerol) with Roche protease and phosphatase inhibitors; cells treated with DMSO, BafA (100 nM), and MG132 (100 uM) for 5 h; lysate clarified at 14,000 x g for 10 min at 4C and quantified by BCA assay. 60 ug lysate loaded; samples denatured in Laemmli buffer with 50 mM DTT at 95C for 5 min; SDS-PAGE run at 120 V; PVDF membrane; wet transfer at 400 mA for 60 min. Blocking in 4% BSA in PBS-T for 1 h RT; primary antibody 1:1000 overnight at 4C; washes in PBS-T; LI-COR fluorescent secondary antibody 926-68071 at 1:10000 for 1 h; visualization on LI-COR.
Dilution: Primary: 1:1000; Secondary: 1:10000
Results Summary: EDEM1 was detected by western blot in WT HAP1 cells after BafA treatment. The blot shows a band around the expected EDEM1 region, with beta-actin included as a loading control.
Comments: Transfer efficiency was not tested by Ponceau S or Coomassie Blue.
