Principle of Assay
This kit provides a simple method for detecting total cholesterol (TC) concentration in a variety of biological samples such as serum, plasma, animal tissues and cells, and bacteria. In the assay, esterase catalyzes the hydrolysis of cholesterol esters to produce free cholesterol (FC) and free fatty acids. Then cholesterol oxidase oxidizes free cholesterol to produce delta-4-cholestenone and hydrogen peroxide. Hydrogen peroxide, 4-aminoantipyrine and phenol react in the presence of peroxidase to form red quinone compounds which have a characteristic absorption peak at 500 nm. The color depth is proportional to the total cholesterol content.
