Antiserum was solid phase adsorbed to ensure class specificity, and this antibody was subsequently isolated by affinity chromatography using antigen coupled to agarose beads. F(ab')2 fragments were generated using a pepsin digestion, and Fc fragments and whole IgG molecules were removed. F(ab')2 fragments were then conjugated to fluorescein isothiocyanate (FITC).
