Recommended Dilutions
In double radial immunodiffusion (Ouchterlony) serum or plasma samples are prepared in serial two-fold dilutions from 1:8 to 1: 128 and placed in the peripheral wells of the rosette arrangement. One well is filled with pooled human serum or another control. The center wells are filled with the subclass specific antiserum and the sera are allowed to diffuse at ambient temperature for 16-24 hours. Frequently the precipitin lines become visible only after staining.
