Principle of Assay
Lyme Disease Borrelia burgdorferi sensu lato IgG + IgM ELISA Kit employs an indirect ELISA for the quantitative measurement of IgG and IgM antibodies to Borrelia burgdorferi sensu stricto, B. afzelii, B. garinii, B. mayonii, B. spielmanii, B. bissetiae, B. bavariensis, and B. valaisiana in serum and plasma. The 96-well microplate is pre-coated with Borrelia antigens. Samples and standards are added and incubated, allowing specific antibodies to bind to the immobilized antigens. After washing, a biotinylated detection antibody is applied, followed by HRP-streptavidin. TMB substrate is added to generate a colourimetric reaction catalyzed by HRP. The reaction is stopped with acidic solution, and absorbance is read at 450 nm. The signal intensity is proportional to the concentration of Borrelia-specific antibodies present and is determined by comparison with a standard curve.
