This antibody binds specifically to the C-terminal region of human C5aR, detecting the non-phosphorylated isoform. It is validated for Western blot analysis, supports immunoprecipitation from cell and tissue lysates, and is suitable for immunohistochemistry in cultured cells and tissue sections.

Confirmation of Complement C5a receptor 1 (C5aR) expression by Western blot in transfected HEK293 cells. Lysates from native HEK293 cells (MOCK) or those stably expressing C5aR were probed with the phosphorylation-independent anti-C5aR antibody (A334503) at a 1:1000 dilution.
Immunocytochemical detection of Complement C5a receptor 1 (C5aR) in HEK293 cells. HEK293 cells stably expressing C5aR were untreated or treated with 1 µM C5a for 30 min and stained with anti-C5aR antibody (A334503) at 1:200. In untreated cells (0 min), C5aR receptors were localized to the plasma membrane, while after 30 min of C5a exposure, receptors appeared in perinuclear vesicular clusters.
Immunohistochemical localization of Complement C5a receptor 1 (C5aR) in human spleen. Spleen sections were dewaxed, microwaved in citric acid, and incubated with anti-C5aR antibody (A334503) at 1:100, followed by biotinylated anti-rabbit IgG and avidin-biotin solution. Color was developed with 3-amino-9-ethylcarbazole (AEC), and sections were counterstained with hematoxylin. C5aR receptors were detected at the plasma membrane of a specific subset of spleen cells.
Identification of Complement C5a receptor 1 (C5aR) in human spleen by immunohistochemistry. Human spleen sections were dewaxed, microwaved in citric acid, and stained with anti-C5aR antibody (A334503) at 1:100. Sections were treated with biotinylated anti-rabbit IgG and avidin-biotin solution, developed with 3-amino-9-ethylcarbazole (AEC), and counterstained with hematoxylin. C5aR receptors were observed at the plasma membrane of a distinct cell population.
Detection of Complement C5a receptor 1 (C5aR) in human lung by immunohistochemistry. Lung sections were dewaxed, microwaved in citric acid, and incubated with anti-C5aR antibody (A334503) at 1:100, followed by biotinylated anti-rabbit IgG and avidin-biotin solution. Color was developed with 3-amino-9-ethylcarbazole (AEC), and sections were counterstained with hematoxylin. C5aR receptors were uniformly detected at the plasma membrane of lung macrophages.
