Human + Mouse + Rat Adalimumab Pharmacokinetic ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of Adalimumab levels in human, mouse and rat serum and plasma.

Direct Sandwich (Quantitative)

Human + Mouse + Rat Adalimumab Pharmacokinetic ELISA Kit is a direct sandwich ELISA that provides quantitative determination of free adalimumab targeting TNF alpha in serum and plasma. The assay employs a 96-well microplate pre-coated with anti-adalimumab capture antibody. Calibrators, quality control samples, and diluted test samples are added to the wells, allowing adalimumab present in biological matrices to bind to the immobilized antibody. After washing to remove unbound components, an enzyme-linked anti-adalimumab detection antibody is added. Following a final wash, substrate solution is added and color development is measured at 450 nm. The intensity of the color is directly proportional to the amount of adalimumab present and concentrations are calculated from the standard curve.

Samples should be collected as serum or plasma using EDTA or heparin as anticoagulants. Samples may be stored at or below -20 °C for up to one year prior to analysis. Before assay, calibrators and test samples should be diluted 1:50 with assay buffer by adding 5 µL of sample or calibrator to 245 µL of assay buffer and mixing thoroughly. Diluted samples should be used immediately and must not be stored.
