Adipogenesis is the process whereby undifferentiated mesenchymal stem cells (MSCs) or preadipocytes develop into mature adipocytes, the specialized fat cells that store energy in the form of triglycerides in adipose tissue. Adipogenesis is initiated by various extracellular signals and transcription factors. One of the key inducers is insulin, which promotes the differentiation of preadipocytes into mature adipocytes. Other signalling molecules, such as growth factors (e.g., insulin-like growth factor 1, IGF-1), cytokines (e.g., tumour necrosis factor-alpha, TNF-α), and hormones (e.g., glucocorticoids), also influence adipogenesis. The key regulators of adipogenesis are a family of transcription factors, including peroxisome proliferator-activated receptor gamma (PPARγ) and CCAAT/enhancer-binding proteins (C/EBPs). These factors control the expression of genes involved in adipocyte differentiation, lipid metabolism, and insulin sensitivity. For example, PPARγ and C/EBPs stimulate the expression of genes involved in the uptake of fatty acids and the synthesis of lipids within MSCs. This includes genes responsible for transporting fatty acids into the cell and enzymes involved in fatty acid synthesis. PPARγ and C/EBPs can also regulate each other's expression, forming a positive feedback loop. They also regulate other transcription factors involved in adipogenesis, such as KLF5 (Kruppel-like factor 5), which further promotes the differentiation of MSCs into adipocytes. Adipogenesis occurs in several stages. First, MSCs or preadipocytes commit to the adipogenic lineage. Then, they undergo clonal expansion to increase cell numbers. Finally, the cells differentiate into mature adipocytes, characterized by the accumulation of lipid droplets. The hallmark of mature adipocytes is the accumulation of lipid droplets filled with triglycerides which are essential for energy storage. However, adipocytes are not simply passive energy storage cells; they also secrete bioactive molecules termed adipokines. These include adiponectin, leptin, and various pro-inflammatory cytokines which play important role in regulating body metabolism, appetite, and inflammation. Adipocytes require functional mitochondria to oxidize fatty acids and generate energy. The adipocyte differentiation process involves the development of an extensive mitochondrial network to support the increased energy demands of mature adipocytes. Adipocytes continually engage in the processes of lipolysis (breakdown of stored triglycerides) and lipogenesis (synthesis of triglycerides). These processes are regulated by hormones like insulin and catecholamines, and they determine the overall lipid content of adipocytes. Adipocytes have a profound impact on whole-body metabolism. They regulate glucose homeostasis by secreting adiponectin, which enhances insulin sensitivity, and they store and release triglycerides in response to energy needs. Hormones such as insulin, glucagon, cortisol, and thyroid hormones play critical roles in modulating adipogenesis and adipocyte function. Insulin, for example, promotes lipid storage in adipocytes, whilst catecholamines (epinephrine and norepinephrine) stimulate lipolysis. Dysregulation of adipogenesis is a central feature of obesity. In obese individuals, adipose tissue undergoes hypertrophy (enlargement of existing adipocytes) and hyperplasia (increased formation of new adipocytes). This contributes to metabolic disturbances and insulin resistance. Finally, certain adipocytes, known as brown and beige adipocytes, are specialized for thermogenesis.
