Chondrogenesis describes the process by which cartilage tissue is formed during embryonic development and subsequently throughout life. Cartilage is a specialized form of connective tissue that provides structural support and mechanical cushioning in various parts of the body, including the joints, nose, and ears. Chondrogenesis initiates with the differentiation of mesenchymal stem cells (MSCs) into chondroprogenitor cells. These progenitor cells express specific transcription factors like Sox9, which initiates the chondrogenic differentiation program. For example, Sox9 regulates the expression of key genes responsible for cartilage formation, such as collagen type II (COL2A1) and aggrecan (ACAN). In addition to Sox9, other transcription factors like Runx2 and Runx3 play important roles in regulating chondrogenesis. Runx2, for example, promotes osteogenesis (bone formation) whilst suppressing chondrogenesis, highlighting the balance between different tissue fates. Chondrogenesis is characterized by the secretion of a specialized ECM. This matrix consists of collagen type II and proteoglycans, primarily aggrecan. Collagen type II provides the structural integrity to the cartilage, whilst aggrecan's hydrophilic nature enables cartilage to retain water, maintaining its mechanical resistance to compression and elasticity. Signalling pathways are involved in the regulation of chondrogenesis, with transforming growth factor-beta (TGF-β) and bone morphogenetic proteins (BMPs) being key players in its regulation. These signalling molecules trigger events that lead to the differentiation and maturation of chondrocytes, the cells responsible for maintaining cartilage throughout life. During chondrogenesis, chondrocytes can undergo hypertrophy, a process in which they enlarge and produce factors including vascular endothelial growth factor (VEGF) and matrix metalloproteinases (MMPs). This hypertrophic cartilage eventually undergoes endochondral ossification, where it is replaced by bone tissue. This process is particularly critical for the longitudinal growth of bones. In addition, chondroprogenitor cells are sensitive to mechanical signals and respond to changes in mechanical loading on cartilage tissue with such forces influencing their differentiation and activity. For example, chondroprogenitor cells express ion channels and mechanosensors that can directly sense mechanical deformation. These mechanosensors include mechanosensitive ion channels (e.g., stretch-activated ion channels) and surface receptors (e.g., integrins or mechanogated ion channels). When these sensors are activated by mechanical forces, they initiate signalling events resulting in the upregulation of genes associated with chondrogenesis and the production of extracellular matrix components, such as collagen and proteoglycans. Cartilage is typically surrounded by a layer of connective tissue called the perichondrium, which contains cells capable of differentiating into chondroblasts. The perichondrium thereby provides a source of new chondrocytes for cartilage maintenance and repair. Chondrocyte maturation involves a transition from proliferating chondrocytes to post-mitotic, hypertrophic chondrocytes. This process is regulated by paracrine signalling, involving factors like Indian hedgehog (IHH) and parathyroid hormone-related protein (PTHrP), which maintain the growth plate's architecture and regulate bone elongation. Cartilage has limited regenerative capacity compared to other tissues. Injury or disease can lead to the degradation of the ECM and to chondrocyte loss. Understanding chondrogenesis is critical in the context of musculoskeletal diseases, such as osteoarthritis and genetic disorders like achondroplasia.
