Transporters

Neurotransmitter transporters are membrane proteins that facilitate the reuptake of neurotransmitters from the synapse back into the presynaptic neuron. This reuptake process terminates the action of neurotransmitters and maintains regulated synaptic signalling. There are several types of neurotransmitter transporters, and each plays a specific role in regulating neurotransmitter levels in the synapse. For example, the serotonin transporter (SERT) is responsible for the reuptake of serotonin. By removing serotonin from the synapse, SERT helps to terminate its signalling and prevent excessive activation of serotonin receptors. Dysregulation of SERT activity has been implicated in mood disorders, such as depression and anxiety, whilst selective serotonin reuptake inhibitors (SSRIs) which block the serotonin transporter and increase serotonin levels in the synapse, may benefit individuals with depression and anxiety. Dopamine Transporter (DAT) is responsible for reuptake of dopamine, a neurotransmitter involved in reward processing, motivation, and movement control. By clearing dopamine from the synapse, DAT helps to regulate dopamine signalling and prevent prolonged activation of dopamine receptors. Dysfunction of DAT is also associated with various neuropsychiatric disorders, including Parkinson's disease and addiction. Norepinephrine Transporter (NET) is responsible for the reuptake of norepinephrine (noradrenaline), a neurotransmitter involved in the fight or flight response, attention, and mood regulation. By removing norepinephrine from the synapse, NET plays a crucial role in terminating its signalling and maintaining homeostasis. Dysregulation of NET has been linked to mood disorders and attention-deficit/hyperactivity disorder (ADHD). Gamma-Aminobutyric Acid Transporters (GATs) are responsible for reuptake of gamma-aminobutyric acid (GABA), the primary inhibitory neurotransmitter in the brain. GATs therefore play a critical role in terminating GABAergic neurotransmission, which is essential for maintaining the balance between excitatory and inhibitory signalling in the brain. Dysfunction of GATs is associated with epilepsy and anxiety disorders. Excitatory Amino Acid Transporters (EAATs) are responsible for reuptake of glutamate, the primary excitatory neurotransmitter in the brain. EAATs help to regulate the concentration of glutamate in the synapse, preventing excessive activation of glutamate receptors, which can lead to excitotoxicity and neuronal damage. Dysfunction of EAATs has also been implicated in neurodegenerative diseases and psychiatric disorders. Vesicular Monoamine Transporters (VMATs) are responsible for packaging monoamine neurotransmitters, such as dopamine, norepinephrine, and serotonin, into synaptic vesicles for subsequent release into the synapse. VMATs are crucial for maintaining proper neurotransmitter storage and release. Dysfunction of VMATs may lead to conditions like Parkinson's disease and certain psychiatric disorders. Similarly, Vesicular Glutamate Transporters (VGLUTs) are responsible for packaging glutamate into synaptic vesicles in glutamatergic neurons. Glutamate is the most abundant excitatory neurotransmitter in the brain, and VGLUTs are essential for proper excitatory neurotransmission. Dysfunction of VGLUTs have also been associated with various neurological and neuropsychiatric disorders. The proper functioning of neurotransmitter transporters is essential for maintaining neurotransmitter homeostasis, synaptic plasticity, and overall brain function. Dysregulation or dysfunction of these transporters can lead to various neurological and psychiatric disorders. For example, altered dopamine transporter function is implicated in conditions like attention-deficit/hyperactivity disorder (ADHD) and Parkinson's disease, whilst dysfunctional serotonin transporters are associated with mood disorders like depression and anxiety.
